A collaboration between the American Dental Association Forsyth Institute and Brigham and Women’s Hospital is pushing the boundaries of periodontal regeneration with a novel approach to bone grafting. Scientists are developing three-dimensional (3D)-printed, patient-specific bone graft materials enhanced with specialized molecules that actively resolve inflammation, offering a smarter, more effective solution for managing bone loss caused by periodontitis.

This innovation stems from decades of research into the body’s immune response, particularly the discovery of specialized pro-resolving mediators (SPMs), natural molecules responsible for signaling the end of inflammation. By incorporating SPMs directly into bioengineered graft materials, the team aims to not only replace lost bone, but also ensure that the healing process is not disrupted by chronic inflammation.

Traditional bone grafts can take 6 months or more to heal, with mixed success rates. In contrast, these customized grafts promise faster integration, less risk of graft failure, and better long-term stability, especially for aging patients whose natural SPM levels decline over time.
